Purification of oncolytic measles virus by cation-exchange chromatography using resin-based stationary phases

چکیده
Cancer therapy with oncolytic measles virus (MV) requires high titers of pure, infectious particles (~108 times more per dose than the vaccine). A gentle but efficient downstream purification process is therefore essential. Here we investigated suitability ion-exchange chromatography for MV using resin-based stationary phases. The success was evaluated based on yield, ratio to total particles, and quantity major contaminants (host cell DNA protein). After testing different anion-exchange, cation-exchange hybrid materials, found that cation-exchanger Eshmuno ® CPX (Merck) most promising resin. By optimizing pH feed buffers, reducing flow rate, achieved a yield 80.7% while by only 16.8%. We also removed 98.3% protein 80.5% host DNA. Although not yet typically used purification, have developed new approach improve efficiency during processing.

Purification of Monoclonal Antibodies Using a Fiber Based Cation-Exchange Stationary Phase: Parameter Determination and Modeling
Monoclonal antibodies (mAb) currently dominate the market for protein therapeutics. Because chromatography unit operations are critical for the purification of therapeutic proteins, the process integration of novel chromatographic stationary phases, driven by the demand for more economic process schemes, is a field of ongoing research. Within this study it was demonstrated that the description ...

Ion-exclusion/cation-exchange chromatography (IEC/CEC) on a weakly acidic cation-exchange resin using on acidic eluent, developed by Tanaka et al. in 1994, has been employed to monitor the quality of environmental water samples such as acid rainwater and river water, to analyze body fluid samples such as saliva, and to evaluate water purification by photocatalytic materials.1–8 One advantage of...
متن کاملCharacters of Kinetic and Equilibrium of Adsorption of Eu(III) by an Cation Exchange Resin
The sorption and desorption behaviors of Eu(III) from aqueous solutions with cation exchange resins containing carboxyl groups (D155 resin) were studied in terms of varied Eu(III) concentration, adsorbent dosage, pH, contact time and temperature. Batch shaking sorption experiments were carried out to evaluate the performance of D155 resin in the removal of Eu(III) from aqu...

متن کاملPurification of prothrombin complex proteins from human plasma in anion exchange resin using pseudoaffinity chromatography
Background Prothrombin complex contains the vitamin K dependent coagulation factors II, VII, IX, X, protein C, and protein S. It has been used for the treatment of congenital coagulation disorders and is recommended for reversing oral anticoagulation [1]. Prothrombin complex proteins require Ca to express their activities. The conformational change induced by Ca finds a practical application in...
